Santander de Quilichao es denominado tierra de oro debido a las explotaciones auríferas que se realizaban a lo largo de algunos de sus ríos, entre los que se destacaba Quinamayo, para lo cual se utilizaban esclavos de la raza negra.
La fundación oficial de Santander de Quilichao ocurrió el 20 de enero de 1878, por parte de un grupo de colonos dirigidos por el colombiano Juan Francisco Salazar. La localidad recibió este nombre en honor al general Francisco de Paula Santander, uno de los próceres de la independencia de Colombia.
